The main collection of the museum was formed in 1921-1931 on the basis of the collections of the Caucasian Ethnographic Society, the museums of antiquities of New Nakhichevan, the ancient fortress of Ani, and the archive of Vagharshapat. In 1923, the Committee for the Protection of Monuments of Antiquities of Armenia was created under the leadership of A. Takhanyan, with the participation of T. Toramanyan, M. Saryan , A. Kalantary, S. Terkobyants, and since 1925 – N. Bunyatyan, B. Hasan-Jalal, G. Arch. Gevorkyan, St. Lisitsyan, and later – E. Shakhaziz, Taragros, K. Gafadaryan, S. Barkhudaryan, N. Tokarska. The committee transferred a significant part of the exhibits to the museum, which replenished its base, forming a collection of 15,289 items.

Currently, the museum holds about 400,000 exhibits of the national collection, divided into sections: archeology (35% of the main collection), ethnography (8%), numismatics (45%), documents (12%). The collections are constantly replenished mainly by finds excavated in ancient monuments on the territory of the Republic of Armenia, as well as purchases and gifts.
